Ashwagandha (Withania somnifera) is classified as an āadaptogenicā herb, indicating an ability to support how the body physiologically adapts. In the Ayurvedic tradition, ashwagandha is considered a rasayana,5 meaning it is used as a rejuvenator to achieve homeostasis, build vitality, and slow degenerative and senescence-related processes. For thousands of years, ashwagandha has been used to help support people with ailments ranging from musculoskeletal conditions to sleep issues. It is widely used today for restoring physical and mental energy while balancing the effects of daily stress. Analysis of ashwagandha has revealed that it contains a wide range of biologically active chemical constituents collectively known as withanolides.
Shoden is an extract derived from the leaves and roots of the ashwagandha plant. This multipatented ashwagandha extract is distinguished by its industry-leading concentration of 35% withanolide glycosides. Shoden has been evaluated for its adaptogenic role in the stress response and for the support of restful sleep.
To ascertain the effect of Shoden on restorative sleep, healthy subjects (N = 150) were given 120 mg of Shoden daily for 6 weeks in a doubleblind, placebo-controlled study. Overall sleep quality was evaluated using a weekly questionnaire and an assessment of parameters, such as sleep latency, sleep efficiency, and total sleep time, as measured by a wearable sleep actigraphy diagnostic monitoring device. A significant improvement in sleep measures that define nonrestorative sleep was noted in the treatment group when compared with the placebo, demonstrating the positive effect of Shoden on encouraging restful sleep.
The mechanistic actions of Shoden and their potential impact on sleep were assessed using in vitro and in vivo animal model experiments. Shoden was found to have calming properties, including involvement in the induction of sleep, GABA-mimetic action, non-REM sleep patterns, and slow-wave electroencephalograph (EEG) cycles, suggesting potential for improving sleep quality.

Emerging evidence indicates that polyphenolic compounds in plants may have benefits for sleep quality. Epigallocatechin gallate (EGCG), a polyphenol compound found in green tea (Camellia sinensis), is well known for its demonstrated antioxidant activity. In addition, EGCG also prolongs the effects of GABA (gamma-aminobutyric acid), an inhibitory neurotransmitter that plays a role in the relaxation of the central nervous system. This occurs via positive allosteric modulation of the GABAA receptor by EGCG, resulting in enhanced GABAA sensitivity.
Rosmarinic acid, found in plants of the Lamiaceae family, such as rosemary, lemon balm, and mint, also influences GABAās neurotransmitter activity through positive allosteric modulation of the GABAA receptor. This results in a mild inhibition of glutamate, the main excitatory neurotransmitter, which may contribute to the effect of rosmarinic acid on sleep quality. In addition, rosmarinic acid from plants, including spearmint (Mentha spicata), has been reported to benefit cognitive performance and attention. An inhibitory effect on the acetylcholinesterase and butyrylcholinesterase enzymes responsible for breaking down acetylcholine, a neurotransmitter that is crucial for cognitive functions such as learning, memory, and attention, is thought to be a contributing factor.
A polyphenol-rich blend of spearmint and green tea extracts, known as Daily Zz, has been studied for its benefit to sleep quality and next-day cognitive function. In a randomized, double-blind study involving 89 subjects with subclinical sleep disturbances, participants who received 485 mg/d of Daily Zz for a 30-day trial period were assessed for improvements in sleep, mood, and neurocognitive function. Ā An improvement was noted in sleep quality, as well as the ability to fall asleep, stay asleep, and wake refreshed, as recorded by subjects in a sleep diary and with an activity tracker.
